Freezability of camel spermatozoa as affected by cryoprotective agants and equilibration periods added with pentoxifylline

Abstract |
Ten sexually mature 6 to 10 years old male dromedary camels of 500 to 600 kg body weight were used in two experiments. The first experiment aimed to define the effect of extended camel semen with Tris-Yolk-Fructose (TYF) extender, different cryoprotectants (2, 4 or 6 % glycerol: GL, dimethylsulfoxide: DMSO and their combinations: GL plus DMSO) and equilibration periods (0, 2, 4 hours) on the percentages of post-thawing motility, freezability and acrosomal damage. The second experiment aimed to define the effect of different concentrations of pentoxyfilline (0, 0.2, 0.4, 0.6, 0.8, 1.0, 2.0, 4.0, 6.0 and 8.0 mM/100ml) on post-thawing motility, freezability, acrosomal status and enzymatic activities during thawing incubation at 37 ˚C for up to 2 hours. Sperm penetration into she-camel cervical mucus during incubation at 37°C for up to 4 hours was also recorded. The results showed that the extended camel semen with TYF extender with 6% DMSO and 4 hours as equilibration period had significantly (P<0.05) improved the percentages of post-thawing motility , freezability and acrosomal damage of camel spermatozoa compared to of GL, DMSO alone or GL plus DMSO ( First Experiment). The second Experiment shows that the addition of 6.0 mM pentoxifylline to the extended camel semen increased significantly (P<0.05) the percentages of post-thaw motile spermatozoa, freezability, acrosomal status, sperm penetration into she-camel cervical mucus and decreased significantly (P<0.05) the amount of aspartate-aminotransferase (AST) and alanine-aminotransferase (ALT) enzymes released into the extracellular medium compared to different concentration of pentoxifylline during thawing-incubation at 37°C for up to 2 hours. The prolongation of thawing-incubation time at 37°C decreased significantly (P<0.05) the percentage of post-thawing motility, freezability and sperm penetration into she-camel cervical mucus while, significantly (P<0.05) increased the percentage of acrosomal status and the amount of AST and ALT enzymes released into the extracellular medium.
