Comparative chicken genome analysis of Egyptian local breeds and developed strains 1. The microsatellite discrimination between Dandrawi and Sinai breeds

Abstract |
This study represents assessment the genetic characterization of two Egyptian chicken populations; Dandrawi native breed and Sinai strain. On the basis of molecular genetic characterization a set of ten highly polymorphic microsatellite markers were utilized. The discriminating power of such microsatellite was estimated. A total of 120 alleles was detected averaged 12/marker with allele frequency ranged from 0.01 to 0.65. The expected heterozygousity (HE) in Dandrawi ranged from 0.16:0.85 and averaged 0.67. With respect to Sinai strain the HE was (0.61:0.86) and averaged 0.73. A total of 32 private alleles (26.67%) were detected indicating its validity in diagnostic and fingerprint as tool for breed identification. It was concluded that this particular set of microsatellite markers; especially, ADL176 (for its highly polymorphism versus all loci, 22 alleles) can be used as a measure of genetic divergence in native chicken breeds and/or strains in Egypt. The results obtained herein confirm a completely genetic distance of two chicken used in this study. In addition, The Sinai strain showed a highly polymorphism than Dandrawi breed. This might be attributed as Dandrawi breed viewed breeding stress more than that of Sinai.
