English : Semen cryopreservation for two local chicken strains as a tool for conservation of Egyptian local genetic resources

Abstract Semen cryopreservation is the most practical method for the long term storage of poultry genetic resources. The objective was to compare two chicken strains (El-Salam and Dokki-4) for cryopreservation of sperm, two extenders (Tris-egg yolk and Egg yolk citrate) and two cryoprotectants (glycerol and dimethyl-acetamide) with three levels (3, 7 and 10%). At the beginning of the trial all birds were 42 weeks of age. Semen was collected twice a week from twenty cocks from each strain and 150 hens (75 per each) from the same strains were used for fertility trails. Semen was collected, diluted (1: 4v/v) and analyzed for motility, livability and abnormality at three times (post dilution, equilibration and frozen-thawed) Results declared that strains had no significant differences for post diluted, post equilibrated and post frozen-thawed traits. Tris egg yolk extender had higher significantly effect on post frozen-thawed motility (29.93±0.53) and livability (31.20±0.52) of spermatozoa than egg yolk citrate extender. The best significant cryoprotectants were 7% Glycerol and 3% DMA for post frozen-thawed motility and livability of spermatozoa for both strains. Tris egg yolk extender had higher significant recovery rate for post frozen-thawed motility and livability. Among cryoprotectant treatments, 7% glycerol followed by 3% DMA achieved significantly highest response in both post frozen-thawed motility recovery and livability for both strain . The fertility rates obtained with frozen and thawed spermatozoa with 3%DMA and 7% glycerol yielded fertility percentage 28.42 and 37.76% for El-Salam strain, respectively. The hatchability rates were 88 and 91.43 % for 3%DMA and 7% glycerol in El-Salam strain, respectively. While, Dokki-4 gave 0 % for fertility and hatchability.
