English : Effect of Melatonin on in vitro development of rabbit embryos recovered from red Baladi breed

Abstract This study aimed to investigate the effect of melatonin supplementation at different levels in culture medium on embryo development in rabbits. Embryos were recovered from nulliparous Red Baladi rabbit does by laparotomy technique 48 hours after insemination. Normal embryos of 16-cell stage were cultured for 3 days in the following four culture medium; (i) control (TCM-199 + 20% fetal bovine serum), (ii) control + 10-3M melatonin, (iii) control + 10-6M melatonin and (iv) control + 10-9 melatonin. The development rate to expanded and hatched blastocyst stages were calculated for each group. It was clearly observed that the high level of melatonin at 10-3 M adversely affected the in vitro development rates. A significant decrease was found in the total development rate of 10-3M group in comparison with other groups (53.9% for 10-3M vs. 88%, 85.6% and 81% for l0-6M, 10-9M and control, respectively, P<0.05). Embryos hatchability was also significantly decreased in 10-3M melatonin in comparison with other groups (29.5% for 10-3M vs. 79.2%, 68.1% and 61.4% for 10-6M, 10-9M and control, respectively, P<0.05). On the other hand, abnormality rate in developed embryos was significantly higher when the culture medium supplemented with 10-3 M melatonin (49.4%) in comparison with l0-6M and to-9M groups (11.9% and 14.4%, respectively, P<0.05). Results show that a moderate level of melatonin (10-6M) may improve the development and hatchability rates of recovered embryos and induce a lower abnormality than that in the control group or physiological level of melatonin (l0-9M), however, these results are not significant and more studies are necessary to prove.
