Plasmid mediated carbapenems resistance in clinical and environmental isolates of pseudomonas aeruginosa recovered from sheep farm

Abstract |
Resistance to carbapenems was observed in all clinical P. aeruginosa isolates and 78.5% of environmental isolates recovered from a sheep farm. Plasmid profile analysis was performed using agarose gel electrophoresis. All the P. aeruginosa isolates were found to harbour a single and similar plasmid 14.5 kb except one soil isolate which harboured plasmid 29.9 kb. Successful transformation of resistant plasmid was done between P. aeruginosa as a donar and E. coli XLI as a recipient by electroporation. It was noted that the transformants E. coli XLl resisted to carbincillin while it was sensitive to all antibiotics prior to the experiments. Agarose gel electrophoresis of the tranformants showed the presence of transforming plasmid 14.5 kb. In conclusion carbapenem resistance P. aeruginosa is of particular importance because it is plasmid mediated and is transferable to gram negative rods. It is suggested the cross transmission of strains as well as spread of plasmid or integron borne gene within the farm. Determination of the reservoris which may be environmental may help to prevent dissemination of carbapenemase gene so plasmid should be characterized in all multidrug resistance P. aeruginosa and nation wide antibiotic policy should be made to minimize the emergence of antibiotic resistance.
