Abstract Four groups, each of 20 chickens were vaccinated with an inactivated egg drop syndrome (EDS) vaccine. The vaccine was produced utilizing 4 variables for virus propagation. These were embryonated duck eggs, duck embryo fibroblast cell culture, chicken embryo fibroblast cell culture and chicken embryo rough cell culture (CER). An initial dose (I/M) of 0.5 ml was given per bird at the age of 2 months, boostered 3 weeks later with the same dose. A 12 weeks period of studying the acquired humoral immune response revealed a great approximation in neutralizing antibody and haemagglutination inhibition titres in different groups. Variability of cell-mediated immune response results was found as of lymphocyte transformation and phagocytic activity tests performed through a one month period post initial vaccination. On the basis of immunity studies, all variables were of approximate results, even though chicken embryo fibroblast cell culture gave the most sustainable titres through 9 passages.
