Abstract An Enzyme-linked immunosorobent assay (ELISA) using a lyophilized BHK cell lysate antigen was used for the rapid detection of Rift valley Fever(RVF) antibodies in caprine and ovine sera. Optimal conditions for antigen concentration, serum, and conjugate dilutions were established by checkerboard titrations. The use of phosphate buffer saline (PBS) gave similar results of carbonate/bi-carbonate buffer as coating buffer. The use of skimmed milk 5% in blocking buffer reduced non-specific reactions and omitted the blocking step saving the time of ELISA technique. In case of indirect ELISA, we can use a serum dilution of (1:20) to detect low level of RVF antibodies without non-specific background. the indirect sandwich ELISA method gave better results than indirect ELISA. The specificity and sensitivity of ELISA were determined by the use of pre-and post vaccination sheep and goat serum samples. The results of modiffied ELISA correlated with those of neutralization test revealing 100% specificity and 92% sensitivity.
