English : Trials for preparation of combined vaccine against fowl cholera, new castle, infectious bronchitis and egg drop syndrome diseases in chickens using montanid ISA50 oil
Abstract Montanid ISA 50 was used as an aduvant for preparation of combined inactivated fowl cholera (FC), Newcastle disease (ND), infectious bronchitis (IB) and egg drop syndrome (EDS) vaccine compared with the same vaccine adjuvanted with paraffin oil. Two groups of susceptible chickens were inoculated separately by first dose of these vaccines at 21 days old followed by second dose after 21 days from the first and the third group was left as non- vaccinated control group. Cellular immunity was evaluated against all of these diseases by using lymphocyte blas-togenesis, phgocytic percentage, phagocytic index and hematological examination by leucocyte differentiation test. Humoral immune response was also evaluated by using indirect hemagglutination test (HA), enzyme linked immunosorbant assay (ELISA) for FC, direct hemagglutination inhibition test (HI) for ND and EDS and serum neutralization test (SNT) and ELISA for IB. The results of the aforementioned tests proved that the two types of adjuvant induced protective titre against all diseases, but Montanide oil adjuvant gave higher antibody level then paraffin oil. Protection tests which were done against virulent FC and ND gave similar results.
