Effect of diluent ph on rabbit semen quality during preservation and artificial insemination

Abstract |
Thirty two sexually mature bucks and one hundred sixty four monoparous does in two sequence parities of New-Zealand White (NZW) rabbits were used in two experiments. Experiment one was planned to evaluate physical quality and enzymatic activity of diluted NZW rabbit semen with tris-yolk lactose as influenced by pH media (6.9; 7.1; 7.3 or 7.5), during preservation at refrigeration temperature (4-6 °C) for up to 3 days or incubated at 37°C for up to 4 hours. In fertility traits (experiment 2), the rabbit does were inseminated artificially by using diluted semen at different pH medium (6.9; 7.1; 7.3 or 7.5) to estimate the effect of diluents-pH on fertility trails.
The results obtained showed that, spermatozoa storage ability and physical semen quality represented by percentages of each of advanced sperm-motility, abnormal and dead spermatozoa and acrosomal damages, as well as sperm enzymatic activity represented by activities of glutamic-oxaloacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT), acid phosphates (ACP) and alkaline phosphates (ALP) and pH values were significantly (P ،ـ0.05)better in the diluted semen with pH 7.3 or 7.5 than in 6.9 or 7.1, during chilled storage at (4-6°C) for up to three days or incubated at 37°C for up to four hours. Sperm penetration into estrous cow cervical mucus increased significantly (P <0.05) in 7.3 or 7.5-pH diluent than in 6.9 or 7.1-pH diluent, during semen preservation at incubation condition (37° C) for up to four hours. Semen quality and values of pH-medium decreased significantly (P <0.05),while sperm enzymatic activity and sperm penetration into estrous cow cervical mucus increased significantly (P<0.05)with progression of time at different preservation temperatures.
Generally, it could be noticed that, all physical semen characteristics and semen enzymatic activity in diluted semen at 6.9 or 7.1-p H were superior significantly (P <0.05) than 7.3 or 7.5 at 0 time of preservation at different temperatures.
Kindling rate, litter size and weight at birth values of rabbit does inseminated artificially by using diluted semen at 7.3 or 7.5-pH were significantly (P <0.05) higher than those inseminated using 6.9 or 7.1-pH diluted semen.
