English : Some Trials to prepare a rabies vaccine in tissue culture

Abstract In this study, trials were made for the propagation of S.A.D. rabies virus vaccinal strain in BHK cells. These virus titre after refreshing by several passage on BHK21 cell line, reach up to 105/0.1ml TCID50 with the appearance of strong C.P.E at 36 hours p.i. By comparison the growth curve on T.C. with that in mice to know the optimal time for the highest virus yield, improved that the highest virus yield happened in between the 36 hours, and 60 hours p.i. on BHK (103-105/0.1ml) respectively where in mice inoculation (107-107.4/0.03 ml). The protection test gave the same result what was call the highest yield of virus in between 36 hours and 60 hours which gave a good protection to the G. pigs after challenged with C.V.S. The main aim of this study was to choice the best method for S.A.D. propagation, the most susceptible cells and the most times which gives virus yield, for using that SAD the most times which gives virus yield, for using that SAD strain as a new tissue culture vaccine to replace the commonly used chicken embryo adapted vaccine and to overcome difficulties encountered its use. In later study, the pathogenicity and immunogenicity of this new adapted strain should be tested. Being safe and potent, it could be used for the production of a new tissue culture vaccine, that could be tried in the prophylaxis of animals after evaluation following the known standardized methods.
