English : The Effect of Aqueous Extract of Nigella Sativa on Mycotic Contaminations and Viability of Frozen Ram Semen
Abstract SUMMARY Ten mature rams (2-3 years age) were used in the present study to detect the effect of supplementation of Nigella Sativa at levels 250 and 300 ug/ml of extended semen with or without adding Lamisil at level of 10 pg/ml ,where free diluted semen from any supplementation was used as control. The extended semen samples were incubated at 37°c, cooled and kept at 5°c. for 72 hours and frozen by using IVM system. Percentages of sperm motility after dilution, motility at 5°c., post thawing motility, sperm abnormality and reduction of motility as well as viability index of post thawing, viability index of semen samples kept at 5°c. and acrosomal defects were recoded. All samples were examined mycologically for the presence of toxogenic fungi and pathogenic yeast. The results showed that supplementation of Nigella Sativa at level 250 ug/ml + Lamisil 10 ng/ml extended semen improved significantly (P <0.05) sperm motility after dilution, motility at 5 c, post thawing sperm motility and post thawing viability index while decreased significantly (P <0.05) sperm abnormality, motility reduction and acrosomal defects than Nigella Sativa at level 300 + 10 ug/ml Lamisil diluted semen. Except viability indices at 5°c. did not differ significantly between both concentrations of Nigella Sativa. All samples were examined mycologically for the presence of toxogenic fungi and pathogenic yeast. The obtained results proved that (Nigella Sativa 250 + 10 ug/m Lamisil) were the antimycotics of choice to be used safely as antimycotic agents in combination with antibiotic regime to control the microbial contamination of frozen ram semen as they overcome the fungal infection during semen processing without interference with semen viability and acrosomal integrity. Also the data declared that Lamisil alone and Nigella Sativa alone has low effect on the pathogenic fungal growth.
