English : Influence of Adding Reduced Glutathione and Glucose to the Fertilization Medium on In Vitro Buffalo Embryo Development
Abstract SUMMARY: Glutathione, an ubiquitous sulfydryl compound found in mammalian cells, is a critical intracellular antioxidants that could protect embryos against oxiradical damage by reactive oxygen species (ROS). The objective of the current study was to investigate the effect of re¬duced glutathione and glucose at various concentrations, added to the fertilization medium, on in vitro fertilization dynamics, embryo development and total amount of intracellular glu¬tathione content. In vitro matured buffalo oocytes were fertilized in vitro in modified Ty-rodc's medium supplemented with various glutathione and glucose concentrations. The current results showed that the addition of reduced glutathione to the fertilization medium at a concentration of 0.5 mM significantly (P<0.05) enhanced the fertilization rate (54.79%), blastocyst development (19.53%) and total intracellular glutathione content (7.84 pmol/ oocyte) compared with that in the IVF control media. High glucose concentration (10 mM) added to fertilization medium resulted in a dras¬tic (P<0.05) effect on in vitro fertilization rate (17.39%), development to the blastocyst stage (0.00%) and total intracellular glutathione content (2.60 pmol/ oocyte). The results also demonstrated that supplementation of fertilization medium with 0.5 mM glutathione could overcome the deleterious effect of high glucose concentration. In conclusion, the present results demonstrate that the addition of 0.5 mM glutathione to the fertilization medium improves in vitro buffalo embryonic development even in the pres¬ence of oxidative stress, possibly by means of scavenging reactive oxygen species.
