Evaluation of Fertilizing Efficiency of Buffalo Spermatozoa in Vitro Subsequent Using Some
Capacitating Agents

Abstract |
SUMMARY:
Generally, mammalian spermatozoa unable to penetrate the mature oocytes to induce fertilization otherwise undergoing capacitation and acrosomal reaction. Recently, many ma¬terials have been investigated to improve the sperm functions. Therefore, the present studies is designed to investigate the effects of some capacitating materials on semen characters, penetration and fertilization in vitro.
Buffalo spermatozoa, washed in S.TALP medium, were incubated for 6 h. in either hep¬arin (10, 20, 50, and 100 pg/ml), caffeine (3, 5 and 10 mg/ml), theophylline (5, 10, 20, and 40 mg/ml) or hypoxanlhine (1,5 and 10 mg/ml). Sperm motility, hyperactivity (vigor, circu¬lar movement and tail beating) and acrosomal status were recorded at 0, 2, 4, 5 and 6 h. post-incubation.
Oocytes, aspirated from ovaries of slaughtered buffalo cows, were matured in vitro in filtrated TCM 199 medium for 24h. at 39C in humidified incubator of 5% Co2. Spermatozoa at the most suitable concentration of the capacitating agents, were incubated with matured oocytes for 24h.
From the present results, treatment with heparin (20u,g/ml), caffeine (5 mg/ml), theo¬phylline (lOmg/ml) and hypoxanthine at (lOmg/ml) increased significantly (P<0.05) sperm motility, hyperactivity and acrosomal reaction (80.6711.37; 1.8710.31 and 58.8718.48, resp.), (79.6712.15; 2.1310.13 and 58.0016.92%, resp.), (83.3312.11, 2.210.31 and 63.1317.76%, resp.) and (83.6712.60, 2.210.31 and 66.6717.40, resp.) when compared to those obtained with control and other treatment. After 5h. incubation, the highest sperm mo¬tility, hyperactivity and acrosomal reaction appeared with hypoxanthine treatment (85.0610.83, 2.7810.15 and 79.67 12.75, resp) when compared to the control (16.6714.41, 0.0+0.0 and 6.3311.45, resp). Only lOmg/ml hypoxanthine significantly (P<0.05) improved in vitro penetration rate and male pronucleus formation (MPN) (51.72%) was significantly increased (P<0.05) compared with control (26.39%) and 20mg/ml heparin (32.76%). Caf¬feine and theophylline treatment enhanced significantly (P<0.05) the in vitro fertilization rate (41.27 and 42.67%, resp.) compared to control (26.39%).
As a conclusion, the use of 10 mg/ml hypoxanthine and 10 mg/ml theophylline for sperm capacitation and in vitro fertilization is the protocol which provides the best results for in vitro fertilization in buffaloes. The efficiency of a capacitating agent is a concentra¬tion-dependent and incubation time-dependent with spermatozoa to induce the highest rates of in vitro penetration and fertilization.
