Influence of Different Energy Substrates on Buffalo Oocyte Fertilization and Embryo

Abstract |
SUMMARY:
A scries of experiments had been carried out to investigate the demand of buffalo IVF and IVP to glucose (G), lactate (L) and pyruvate (P) individually or in combination. Ovaries were collected from local abattoirs. Cumulus oocyte complexes (COCs) were aspirated, se¬lected and incubated in M-199 supplemented with 10% FCS, LH (lOug/ml), FSH (5ug/ml) and estradiol (lpg/ml), for 24h in humidified 5% CO2 incubator at 39°C to achieve maturation. Full expanded COCs were inseminated with heparin (20u.g/ml) treated 2x10° sperm/ml and incubated in the basic medium (Tyroid's containing 6mg/ml BSA) {TA} for fertilization. It was supplemented with glucose alone or in combination with lactate and/or pyruvate (Experiment 1) or in glucose free TA containing L and/or P (Experiment 2). The obtained results indicated the highest (P<0.05) penetration and fertilization rates in G.TALP (63.46 and 38.46%, resp.) compared to G.TA (control) (39.58 and 18.75%, resp.). Mean¬while, there were no differences between G.TAL and G.TAP supplemented Fertilization (F) medium (Exp. 1). In experiment 2, glucose free TA supplemented with either L. and/or P. was used. The mature oocytes were distributed into 4 types of F. media either TALP, TAL, TAP or TA (control). The data revealed that, all media supported penetration rate equally while the TALP medium got the highest (P<0.05) fertilization rate 48.61% when compared to the control (30.30%). Also, L. or P. individually supported fertilization rate similarity (42.03% Vs 37.10%, resp.) On the other hand, the effect of glucose containing (Exp. 3) or free (Exp. 4) HEPES-M-199 (HM) supplemented with either L and/or P on embryo develop¬ment in vitro was investigated. The inseminated oocytes were distributed in HM (Exp. 3), without protein source for 48h, drops containing G. LP, G.L, G.P or G. only (control). The highest (P<0.05) cleavage, morula and blastocyst rates were observed in G.LP (56.25, 22.92 and 20.83 %. resp.) compared to control (30.61, 8.16 and 4.08%. resp.). In exp. 4. No signif¬icant differences among treatments in cleavage rate was noticed while superior (P<0.05) morula (16.67%) and blastocyst (11.90%) were recorded in HMLP compared to control (2.22 and 0.00%, resp.).
In conclusion, glucose individually has an inferior effect on penetration and fertilization rates and early embryo support, meanwhile the combination of G, L and/or P support equal¬ly the penetration and fertilization rates. Combination of L and P had a superior effect in penetration and fertilization rates. A mixture of G.LP had the highest support of cleavage, early embryo development up to morula. In addition, combination of LP had a superior ef¬fect in cleavage, morula and blastocyst development than L or P individually or when they omitted from the basic medium.
