English : Studies on Immunnogenicity of different fractions of Pasteurella multocida strains

Abstract The goal of this study is to evaluate the protective value of different bacterial fractions of Pasteurella multocida strains (A and D). These bacterial fractions were extracted by different four methods in comparison with oil adjuvanted vaccine and their immunogeneicty were evaluated by challenge exposure in terms of logs protection, sound immunity to homologous challenge with virulent P.multocida serotype A and D. These prepared fractions ( Capsular extract, O-specific polysaccharide, cell free filtrate, sonicated, extract) and bacterin were injected subcutaneously in rabbits adjuvanted to incomplete Fruend’s adjuvant. The immunizing efficacy of these vaccines was evaluated by indirect haemagglutination test (IHA) as well as enzyme linked immunosorbent assay (ELISA) and challenge exposure. All the prepared vaccines stimulated a marked increase in the haemagglutination geometric mean titres and ELISA mean absorbance values in sera of rabbits and produced a sound immunity in mice and rabbits after challenge exposure. Our results revealed that sonicated extract were better that capsular extracts and oil adjuvanted vaccines and conferred 100% protection against homologous challenge for serotype A . While adjuvanted sonicated extract for serotype D is the best vaccine conferred 100% protection.
