| Abstract |
Onion (Allium cepa L.) plants showing virus like symptoms similar to those caused by onion yellow dwarf virus (OYDV) were observed in the field, Fayoum region. Virus was isolated and identified according to diagnostic hosts, virus stability, aphid transmission, and serological tests (Ouchterlony double diffusion and ELISA tests).
OYDV was found to infect a few number of plants belong to four families. The obtained isolate was found to be transmitted mechanically and by Aphis gossypii insect in a non persistent manner. Stability of the virus was
dilution end point 104° thermal inactivation point 75C and lost infectivity after 3 days. Serological tests (Ouchterlony double diffusion and ELISA tests) were used for rapid detection of the virus.
OYDV caused severe disease of onion plants which incited stunting and streak and leaves malformation which induced yield losses. The percent of weight reduction of infected bulbs compared with healthy bulbs was 49.8 and 59.28% for Giza 20 var. in 2003/2004 and 2004/2005 season, respectively. While was 35.04 and 32.78% for Behiery in the same seasons, respectively. Field observations in Fayoum region revealed that, onion plants infested with Aphis gossypii Glov and Myzus persicae (Sulzer). The presence of the second species was occasionally and was in little numbers. The total number of A. gossypii was higher on Behiery var. than Giza 20 var. being 126 and 109 in 200312004 season and 141 and 117 in 2004/2005 season, respectively. The infection with OYDV was low in beginning of the season and increased accumulatively until the end of the season and ranged from 2 to 14%. The Behiery var. was more susceptible than Giza 20 to OYDV infection.
