Semen quality, enzymatic activities and penetrating ability of spermatozoa into SHE-Camel cervical mucus as affected by caffeine addition

Abstract |
Twelve sexually mature 6 to 10 years old male dromedary camels of 500 to 600 kg body weight, were used to define the effects of various concentrations of caffeine (0, 2.5, 5 and 10 mM/100 ml) addition to the extended camel semen with two different extenders (tris-yolk fructose and lactose-yolk citrate) on the percentage sperm motility and acrosomal damage., during incubation at 37'C for up to 6 hours. Glutamic-oxaloacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT), alkaline phosphatase (ALP) and acid phosphatase (ACP) concentrations were estimated. The penetrating ability of spermatozoa into she-camel cervical mucus was also assessed.
The results showed that the extended camel semen with lactose-yolk citrate had insignificantly higher the percentage of sperm motility and insignificantly lower the percentage of acrosomal damage than tris-yolk fructose extender. The extended camel semen with lactose-yolk citrate had insignificantly lower the amount of GOT and ACP and significantly (p<0.01) lower the amount of GPT and ALP released into the extracellular medium than tris-yolk fructose extender. The 10 mM caffeine added to the extended camel semen increased the percentage of motile spermatozoa significantly (p<0.01) and decreased the percentage of acrosomal damage significantly (p<0.01), amount of GOT, GPT and ALP and decreased the amount of ACP released into the extracellular medium insignificantly. Incubation time at 37°C decreased the percentage of motile spermatozoa significantly (p<0.01), while increased the percentage of acrosomal damage and the amount of GOT, GPT, ALP and ACP released into the extracellular medium significantly (p<0.01). The penetrating ability of spermatozoa improved insignificantly when added with 10 mM caffeine into she-camel cervical mucus.
