Molecular characterization of manhiemia haemolytica extracts and untility in serodiagnosis of pasteurellosis in egypt

Abstract |
Virulence factors of M. haemolytica (A2 ) and P . trehalosi (T4) local isolatcs of caprine origin, represented by outer membrane proteins (OMP ), lipopolysaccharides (LPS), leuko- toxin (LKt ) and bacterial cell lysate (BCL) as well as OMP of P.multocida (B6) standard strain, werecomparativcly analysed for their antigenic structure and cross-reactive components , using sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western immunoblotting . Main differences among M.heamolytica and Pasteurella species (P.trehalosi and P.multocida) were observed in their OMP extracts : A 37 kilo Daltons (kda) antigenic determinate was a M. haemolytica (A2) specific immunoreactive protein subunit. The most unigue croos-reacting antigens in OMPs of P.trehalosui (T4) and P.multocida (B6) versus polyclonal anti- M.haemolytica (A2) antiserum were at 66, 44, 43 and 41 Kda that could be potential Pasteurella related surface antigens. Tha major immunodetected antigenic determinants in all M.haemolytica (A2) extracts analyzed were at 100 – 105, 66 – 68, 41 – 44, 28 – 29 and 16 – 18 Kda. Neither SDS – PAGE nor Western immunoblotting analyses revealed marked differences in LPS core, Lkt and BCL electrophoretic and immuno – reaction patterns between M.haemolytica and P.trehalosi local isolates. In the indirect enzyme - linked immunosorbent assay (ELISA), indirect haemagglutination (IHA) and rapid plate agglutination (RPA) tests, all antigenic extracts of M.haemolytica (A2) were proved effective potential candidates for serovar – wide detection of anti – M.haemolytica antibody in livestock sera (cattle, buffalo, sheep and camel). However, BCL antigen scored higher values than did the OMP then LKt and LPS. The present work extends the epidemiological analyses related to pasteurellosis in Egypt and initiates a potential for development of rapid diagnostic reagents for livestock animals at risk.
