English : Study on the effect of colicin on the growth of "BHK" 21 cells and its susceptibility of FMD virus used in vaccine production

Abstract The present work was carried out to investigate the effect of colicin E1 on the growth of BHK21 cell culture used for FMD vaccine production and accordingly its effect on the immunogenicity of the resulted virus. The egyptian strain of FMD virus (O1/1993) was serially passaged in 9 serial passages of BHK21 cell culture propagated in cell culture media using 3 different supplement (10% newborn calf serum, 10% newborn calf serum and colicin E1 1 unit protein/ml and 10% newborn calf serum and colicin E1 10 unit protein/ml. The obtained virus titer from the cells supplemented with newborn calf serum was 8.55 Log10 CCID50/ml while that obtained with other supplements was 9.0 Log10 CCID50/ml. aal obtained virus harvests showed CF activity of 1/16 by the 9th passages. Three experimental inactivatted vaccine batches were prepared from the obtained virus and inoculated in 3 different groups of guinea pigs. All vaccinated animals did not show any signs of illness post challenge and exhibited good levels of similar values of antibodies as estimated by SNT and ELISA.
