English : Enzymatic activity and fertilizing ability of rabbit semen supplemented with nigella sativa extraction
Abstract Thirty-six sexually mature Californian rabbit bucks (12 months age) were used in the present work. Semen was collected artificially, evaluated, pooled and extended with lactose-yolk citrate extender. Nigella Sativa extraction was added to the extended semen at levels of 0, 50, 100, 150, 200 or 250 µm/ ml. The extended semen was stored at refrigeration temperature (4 - 6 0C) for up to three days or incubated at 37 0C for up to four hours. The present work included two experiments. The first experiment was designed to evaluate the activities of glutamic-oxaloacetic transaminase (GOT); glutamic-pyruvic transaminase (GPT); acid phosphatase (ACP) and alkaline phosphatase (ALP) enzymes in the extended Californian rabbit semen, during preservation at different temperatures as affected by adding different levels of Nigella Sativa extraction. In experiment 2, sperm penetration into cervical mucus influenced by Nigella Sativa supplementation to the extended semen, during incubation condition was tested. Results obtained showed a significant decrease (P≤ 0.05) in values of each of GOT; GPT; ACP and ALP enzymes in extended Californian rabbit semen, during preservation at refrigeration or incubation temperatures due to Nigella Sativa supplementation. The values of GOT; GPT; ACP and ALP enzymes in extended Californian rabbit semen were arranged significantly (P≤ 0.05) descendingly as recorded by adding 0; 50; 100; 150 then (200 and 250) µm/ ml Nigella Sativa, respectively. These enzymes were increased significantly (P≤ 0.05) by advancement of semen preservation at either refrigeration or incubation temperatures. Sperm penetration into cervical mucus during incubation of the extended Californian rabbit semen at 37 0C increased significantly (P≤ 0.05) due to Nigella Sativa supplementation and advancement of preservation time.
