Abstract |
This work was carried out in IVF laboratory (International Livestock Management and Training Center) Sakha, Kafr El-Sheikh Governorate, Animal Production Research Institute, Agricultural Research Center, Ministry of Agriculture. The present work comprised two main experiments. The first one dealt to investigate the maturation, fertilization and development of rabbit oocytes in vitro as affected by season of the year, type of culture medium and level of osmolarity. The 28 female NZW rabbits aged 6-7 months old were divided into two equal groups, 1st group used without any treatment and 2nd group injected with 75 IU PMSG hormones, slaughtered after 60 hr of treatment. Oocytes recovered from 1st and 2nd group were cultured in TCM-199 medium supplemented with FCS or BSA for 20 hr. Oocytes matured in vitro pooled and incubated with capacitated spermatozoa in vitro for 24 hr in Brackett’s or TCM-199 media with 285, 305 or 325 mOsm/kg at different seasons and observed carefully at 24 hr intervals for progress of development up to 6 days. Results reveled that season of the year had a significant effect (P<0.001 or 0.05) on ovaries weights, number of CL and number of visible follicles for the 1st group only. Addition FCS to TCM-199 medium elevated the percentage of oocytes matured in vitro than those of BSA. PMSG treatment increased significantly (P<0.05) the percentage of oocytes maturation. Brackett’s medium elevated the percentage of fertilized oocytes in vitro compared with TCM-199. Fertilization rate was significantly (P<0.05) higher for the oocytes cultured in media of 305 mOsm/kg than those cultured in media of 285 or 325 mOsm/kg. In TCM-199 culture medium increased fertilized oocytes developed to morula and early blastocyst stages compared with Brackett’s medium. Fertilized oocytes cultured in media of 285 or 325 mOsm/kg failed to reach morula and blastocyst stages.
The second experiment 14 female rabbits injected with 150 IU PMSG hormone, 83 hrs later were injected with 75 IU HCG to induce superovulation and divided into two groups. The 1st group was slaughtered after 10 hrs of HCG injection, while the 2nd group was slaughtered after 12 hrs of HCG injection. Ovarian follicular oocytes collected from 1st group and oviductal ova and follicular oocytes collected from 2nd group were inseminated in vitro in Brackett’s or TCM-199 media with capacitated sperm in vitro and incubated for 24 hrs. The fertilized ova were cultured in Brackett’s or TCM-199 media with 305 mOsm/kg. Results showed that in 1st group, there were no significant differences in fertilization rate between Brackett’s and TCM-199 media, however, in 2nd group, Brackett’s medium improved significantly (P<0.05) fertilization rate than TCM-199 medium. Also, in 2nd group, the percentage of fertilization of oviductal ova was significantly (P<0.05) higher than those of ovarian follicular oocytes. In 1st group, TCM-199 medium enhanced embryo development to morula, blastocyst and expanding blastocyst stages compared with Brackett’s medium. In 2nd group, early blastocyst, blastocyst and expanding blastocyst % in TCM-199 medium were slightly higher than those in Brackett’s medium. Also, in 2nd group, oviductal ova fertilized in vitro that developed to morula, early blastocyst and blastocyst% stages were significantly (P<0.05) higher than those from fertilized ovarian follicular oocytes.
