Abstract |
At present, the somatic cloning technology, recently achieved in mammals, permits really novel solutions for biodiversity conservation, foundation of new productive strains as well as the production of transgenic animals.
The two first indicated applications require for techniques of somatic cells cryopreservation, which should be efficient enough to allow to establish a cryobank of nuclear donor cells.
The present Thesis has been entirely performed in the rabbit species and it is chiefly focused on the required techniques to cryopreserve, cultivate and prepare the two main cellular types, actually considered as better nuclear donor candidates for somatic cloning, either for biodiversity conservation or genetic improvement: cumulus cells and especially, epithelial fibroblasts.
Two procedures of non-equilibrium cryopreservation (rapid-freezing and vitrification) have been tested to conserve these two cellular types, reaching very high survival and cellular proliferation rates after warming, and being rather efficient to cover the applied objective pursued here.
Moreover, we have established temporal postmortem limits and environmental temperatures at which, we can recuperate alive cells from dead specimens.
Finally, the different technical steps of somatic cloning have been also studied. Thus, we have grown on our knowledge of the main critical phases, which allows us to suggest possible technical solutions. Moreover, I would like to emphasize the technical efficiency achieved, evaluated as the percentage of cleaved embryos per transferred nucleus, being similar to those reported by other authors in this same species.
