Abstract |
Clostridium perfringens type A enterotoxin was purified by precipitation from the extract of sonically treated cells at 40% saturation of ammonium sulphate to concentrate the enterotoxin and then 15% saturation then by gel filtration on sephadex G-200.
The enterotoxin appeared to be homogeneous on 8% polyacrylamide gel electrophoresis after these three-steps of purification procedure. The mouse intravenous minimal lethal dose (MLD) was 20 MLD/ml after 15% saturation of ammonium sulphate and 80 MID/ml after gel filtration chromatography.
A specific enterotoxin antiserum was obtained by inoculating a rabbit with the active fraction at (32.147 kDa) after gel chromatography on Sephadex G200. This serum neutralize of the enterotoxin and gave one precipitation band with the purified enterotoxin in counter-immunoelectrophoresis.
