Abstract Current methodologies for detecting and identifying Mycoplasma bovis are time consuming and difficult. In the present work, cultural procedure and polymerase chain reaction (PCR) technique were compared. PCR assay was superior to cultivation method (30.55% and 26.55 respectively). DNA amplification using the PCR technique was evaluated for rapid identification of Mycoplasma bovis. Two oligonucleotide primers of 24 and 25 bases in length were constructed (based on RNA genes). The amplification reaction produced a single product of 360 bp in size which was readily detected by agarose gel electrophoresis and ethidium bromide staining This technique provides a simple and extremely sensitive method for the detection of M. bovis in milk and respiratory tract of infected cattle.
