Comparison between different specific media for the isolation and identification of Mycobacterium species

Abstract |
Defenitive diagnosis of mycobacterial disease depending upon isolation and identification of the etiologic agent. Several weeks of incubation required for isolation of mycobacteria on conventional media. The Septi-Chek AFB system was compared with the conventional media (Lowenstien Jensen media) for the detection and isolation of the mycobacteria in 60 specimens from positive tuberclin cows. A total of 45/60 lymph nodes proved to be positive on Septi-Chek AFB system for mycobacterial cultures, 9 were found to be rapid growers and 36 were slow growers, on the other hand 40/60 lymph nodes were detected on conventional media with pyrovate and with 5% Nacl. Six of which were classified as rapid growers. The other 30 isolates were found to be slow growers, four of positive cultures were contaminated on conventional media. The Septi-Chek system showed more reliability and has detected a total of 5 isolates that were not grown on the conventional media and were demonstrated as negative cultures. The mean time for the detection of the slow grower M. bovis and M. intercellularis was 20 days and 18 days for M. tuberculosis, M.tuberculosis, m.kansasi and M. scrofulaceum, while on conventional media, it was 46 days, 36 days and 48 days respectively.
The rapid growers M.phlei and M. smegmatis were detected on the 5th day post inoculation onto Septi-Chek media, while on conventional media, it was 5 days and 7 days for growth respectively. The present study indicates that the Septi-Chek AFB system is rapid sensitive, non automated system for detection of mycobacteria and it is efficient methods to be used in veterinary mycobacterial laboratories
