English : Histological and chemical studies on the activity of the Mammary gland in buffalo at successive stages of season of lactation

Abstract This work was carried out in Animal Production Research Institute Agriculture Research Center, Ministry of Agriculture and Animal Production Department, Faculty of Agriculture, Cairo university, Egypt, throughout 1991-1992. This work was carried out to find the variations in histological and cytological feature of the mammary tissue and chemichal constitulion of milk alongsid blood plasma precursors in buffaloes at successive stages of lactation. The study was carried out data on 15 buffaloes (1-5 lactation ) 3 in each season, the tissue samples, milk and plasma taken at 7d., 35d., 5m. of lactation and at the end of lactation. Tissue samples were taken by sergical method (approxmality 3cm)the biopsy was into two parts, one part was used for light microscopic studies , while the other was applied for electrone microscope cytological study. The milk and blood samples were applied to determine fatty acid in the range C4 - C20 . Fat, protein and lactose percentage were determine in milk samples. Also the total concentration of free amino acids was determined in blood samples. The main results obtained are summarized as follows:- 1- Histological Study:- 1. The higher value of gglandulur tissue percentage was at the peak (35d.) of lactation (84.48%), and lower value at the end of lactation (19.83%). 2. The highest value of alveolus volume was at 35d. (60.81 µ3) and the lower at the end of lactation (8.37 µ3). 3. Number of alveoli in 1 cm3 of tissue was higher in early stage of lactation (7d.). 4. The highest mean value of hight of epithelial cell was at 35d.(9.21 + 0.16 M). 5. Diameter of fat cell was higher at the end of lactation: 82.64 + 10.59, 61.34 + 8.32 for long and short diameter of ovoid shape, respectivelly. 2- Cytological Study:- 1. maximum sub cellular organelles (Endoplasmic reticulum, Golgi bodies and Mitochondria) development occurred at the peak stage of lactation (35d.). 2. The greatest density of secretory products (protein granules and fat dropelts) within the cell was found at the peak stage (7 -35d. of lactation). 3. The large number of lysosomes were showen at the end of lactation ( degenerated cells) . 3- Milk yield and constitution:- 1) Most animals reached the peak of milk secretion at 35d. of lactation, however some animals reached the peak at 7d. of lactation. 2) The individual variation was high between animals in protein and fat percentage. The highest fat percentage was showe at the end of lactation. 3) The total amino acids in blood plasma was decreased at the peak -stage (35d.) with increases of milk yeild. 4) The individual variation was highly cleare between animals for the fatty acids (C6 - C2o) and its percentage at successive stages of lactation.
