Pathological and physiological studies on some isolates of Macrophpmina phaseolina

Abstract |
Eleven isolates of Macrophomina phaseolina isolated from different location in Egypt Governorates and hosts were found to be grow on PDA medium and showed significantly differences in most growth characters. M. phaseolina isolates El-Fayoum- M2, El-Menia- M3, Domiat-M5, Sohag-M4, isolate Qalubia-M10 and isolate Asuite- M8 were more effective for growth on PDA medium than isolate Beni-Suif- M7. Meanwhile, isolates Qalubia-M10, isolate Asuite- M8 and isolate Domiat-M5 were more effective for increasing of sclerotial formation/plat than isolate sesame from Sohag-M4. Concerning the aerial hypha and the visual colour of the growth, results indicated that, all isolates of M. phaseolina showed positively for aerial hypha and the gray to black gray colour but only exception isolate Giza-M1 and isolate Sohag-M4 that showed white colour. The effects of culture filtrate of different isolates of M. phaseolina under two sterilization methods on sesame shoot and root length were study. It clears that, hot sterilization more effective for decreasing of shoot, root length and seed germination % than cold sterilization.
Most of isolates M. phaseolina were found to be pathogenic on sesame plants and showed significant differences between them for the symptoms of pre-, post- emergence damping off and charcoal rot. Also, these isolates were more active for incidence of charcoal rot than pre and post emergence damping off. Results show that, M3, M8 and M10 showed highest aggressive on sesame plants. Regarding the charcoal rot disease on sesame, results show that, M1 to M4 showed height level for charcoal rot incidence, while M5 and M9 were the least effective in this respect.
Concerning the stem that infested by tooth peak method with different isolates of M. phaseolina on charcoal-rot disease incidence on Giza 32 sesame cultivar after 30, 60 and 90 days from sown. Disease incidence % was exhibited significant increases after 90 and 60 days from sowing when compared with the first month after sowing. According the host range results indicated that, all isolates of M. phaseolina were pathogenic on all oil crops that used in this study.
Also, the relationship between the oil crops and M. phaseolina isolates were studied, results revealed that, cotton and soybean were susceptible for all M. phaseolina isolates for pre emergence damping off %. While, sesame and cotton gave the highly level for % post emergence. Charcoal rot % was also measured results indicated that, sunflower was the most effective by these isolates and gave the highly percent of charcoal rot followed by sesame.
